Mesenchymal Stromal Cells

Full-length Biolaminin 521 substrate enables biologically relevant and standardized Mesenchymal Stromal Cells culture.

Standardized and Efficient MSC Culture with
Full-Length Laminin

Mesenchymal stromal cells (mesenchymal stem cells, MSCs) are adherent cells widely studied for their immunomodulatory and regenerative potential, with ongoing clinical studies in graft-versus-host disease, aging-related disorders, autoimmune diseases, and tissue repair. However, variability introduced by undefined and xenogenic culture conditions limits reproducibility and delays clinical translation.

MSCs sense and respond to microenvironmental cues, with extracellular matrix (ECM) interactions playing a central role in regulating identity and function. Laminins, key components of the ECM, are fundamental regulators of cell adhesion, phenotype, and migration within both stem cell and tissue-specific niches.

Biolaminin® substrates are full-length, recombinant human laminins that are xeno-free and chemically defined, providing a standardized, biologically relevant surface for MSCs expansion and differentiation. By mimicking native ECM signals, Biolaminin supports consistent cell behavior across basic research and clinical manufacturing workflows.

Generation, expansion, and differentiation of MSCs using Biolaminin 521. MSCs can be isolated from various tissues or generated from iPSCs as iMSCs on Biolaminin 521. The substrate supports MSC self-renewal, maintenance, and differentiation into adipocytes, osteoblasts, and chondrocytes.

Biolaminin Key Advantages for MSC Culture

Biolaminin 521 supports fast and stable proliferation of both primary MSCs and iMSCs, reducing the time required between passages. The biologically relevant microenvironment enables consistent cell expansion that supports robust and scalable culture workflows.

Full-length Biolaminin supports serum-free culture by providing essential extracellular matrix (ECM) signals that recreate a native-like microenvironment in vitro. By promoting robust cell adhesion and survival, Biolaminin eliminates the need for undefined serum components. Serum-free, chemically defined systems reduce biological variability, improve reproducibility, and minimize batch-to-batch inconsistencies associated with serum-based culture. In addition, animal-origin-free and batch-consistent conditions are critical for meeting regulatory standards and enable the production of clinical-grade cell products for therapeutic applications.

Laminin-521 is the natural laminin isoform present in MSC niches in vivo. Biolaminin 521 recreates this supportive environment in vitro, helping preserve and improve MSC identity and enabling long-term proliferation.

Biolaminin 521 enables scalable MSCs expansion with uniform cell attachment and sustained high proliferation across different surface materials and systems.

Multiple high-quality scientific publications have validated the benefits of full-length laminins to support MSC culture and differentiation. Examples include studies by Tian et al. (2022), Danalache et al. (2024), and Jiang et al. (2017).

iMSCs: Expanding MSCs Sources Through iPSC Technology

Pluripotent stem cell-derived mesenchymal stem cells (iMSCs) offer strong potential for regenerative medicine. iMSCs are generated by differentiating induced pluripotent stem cells (iPSCs) into MSCs, offering new solutions to overcome limitations associated with primary tissue sources and patient variability.

Example of Biolaminin 521 application for the generation and differentiation of iMSCs.
